Effect of Eugenol on High-Fat Diet-Induced Obesity and Depression in Mice

Abstract

|
The excessive intake of a high-fat diet (HFD) leads to psychiatric disorders with obesity through gut dysbiosis. In the present study, we examined whether eugenol could alleviate obesity, depression, cognitive impairment, with neuroinflammation. Long-term exposure to high-fat diet induced depression and cognitive impairment with gut dysbiosis. However, oral administration of eugenol alleviated HFD-induced depression, but not obesity, and suppressed hippocampal tumor necrosis factor (TNF)-¥á expression in the hippocampus and colon-shortening and myeloperoxidase activity in the colon. Furthermore, eugenol treatment suppressed HFD-induced Proteobacteria and Actinobacteria populations in the gut microbiota. Eugenol increased corticosterone-suppressed brain-derived neurotropic factor (BDNF) expression in SH-SY5Y cells and decreased lipopolysaccharide-induced NF-¥êB activation in BV2 cells. These results suggest that eugenol may alleviate depression/anxiety by inducing gut microbiota-involved NF-¥êB activation-suppressed BDNF expression.
